Mutation in the
-1,3-galactosyltranferase gene of the pig
In the current project we are searching for a pig, which carries a genetic modification in the
-1,3-galactosyltranferase gene and because of this could be of great importance in the application of Xenotransplantation.
Following is a step-by-step-description of the screening procedure:

1. Preparation of the DNA from collected tissue-samples
The most effective method to gain tissue samples is the TypiFix-extraction system of AGROBIOGEN. For isolation of DNA DNAFIX 96 columns were used, which are superior to other systems in regard to time of preparation and cost

2. Amplification of target gene fragment
By a nested two-step PCR reaction a 729 bp fragment of exon 9 is amplified. The PCR-product is controlled via gel electrophoresis.
